单层立方上皮为单层扁平上皮及单层柱状上皮的中间型。由单层立方上皮细胞所构成。高立方状与低柱状之间的差别通常是主观的，且只有描述上的价値而已。在以对基底膜做垂直方向的切面时，因为上皮细胞呈现四方形，故传统上将其描述为立方上皮；然而，若从表面上来看，单层立方上皮通常内衬在小的导管和管道上，可能具有排泄、分泌或吸收等功能。肾脏、唾液腺和胰脏的小收集导管便是此例。肾臓小型集尿小管之内衬细胞的细胞界限虽然不明显，但细胞核的形状可约略提供我们细胞大小和形状的判断依据。这些细胞的高度代谢活性反应在其细胞核染色质较散布。且核仁很明显。
