DNA连接酶（EC6.5.1.1），也称DNA黏合酶，在分子生物学中扮演一个既特殊又关键的角色，那就是把两条DNA黏合成一条。无论是双股或是单股DNA的黏合，DNA连接酶都可以借由形成磷酸双脂键将DNA在3'端的尾端与5'端的前端连在一起。虽然在细胞内也有其他的蛋白质，例如像是DNA聚合酶在其中一股DNA为模板的情况下，将另一边的DNA单股断裂端，透过聚合反应的过程形成磷酸双脂键（phosphodiester bond）来黏合DNA。但是DNA聚合酶的黏合过程却只是聚合反应一个附带的功能而已，真正在细胞内扮演DNA黏合反应的工作还是以DNA连接酶为主。。

In molecular biology, DNA ligase is a specific type of enzyme, a ligase, (EC6.5.1.1) that facilitates the joining of DNA strands together by catalyzing the formation of a phosphodiester bond. It plays a role in repairing single-strand breaks in duplex DNA in living organisms, but some forms (such as DNA ligase IV) may specifically repair double-strand breaks (i.e. a break in both complementary strands of DNA). Single-strand breaks are repaired by DNA ligase using the complementary strand of the double helix as a template, with DNA ligase creating the final phosphodiester bond to fully repair the DNA.
